Do you only have TS installed or a big library of games?

From the manual for the Class 421:
1) Locate where you have downloaded this pack and unzip it. Information on how to do this can be found here.
2) Go to the location where you have extracted the files from the .zip file.
3) Now find the .exe file which should look something like this and is called ‘Class 421-422-423 Sound Pack’. Double-click this file.
4) Follow the steps and by the end of the process, this sound pack will have automatically installed to the RailSimulator.com Class 421, RailSimulator.com Southern Class 421, RailSimulator.com Class 422 & RailSimulator.com Class 423.
and answer that I got on UKTS to a similar question to yours was:
"From a quick look at the files, the 421/2/3 pack replaces the various ***Sound.bin files with AP versions, which then call the sounds in the Assets\AP\C421\Audio\RailVehicles\ folder. So, for the Class 423, the RSC version of Assets\RSC\Class423Pack01\Audio\RailVehicles\Electric\Class423\DTCL\DTCL Sound.bin is replaced by the AP version."

There can sometimes be an issue where the AP installer(s) install the program into where it thinks your railworks installation exists rather than where it actually resides.
Sometimes, for whatever reason, a move in location of the railworks folder doesn’t actually register and the only way around it is to manually change the install path every time you install an AP product. It literally triples the install time.
Read his/her first post he quite clearly says the AP SP installs to the Assets\AP folder if it does that then it will be applied to the RSC versions in that same Assets folder. That is how AP installers work. Yes you may have to set the correct path manually in some cases but it cannot affect the install time. How?
Unless of course he/she has two \Railworks\Assets folders due to moving from the C to F drives.
He/She also states quite clearly that the JT registry fix did not work.
My post is merely an explanation in that you may not see changes in the RSC audio files but the AP_sound.bin file takes over from the default, and have installed as they should have done.

I wonder if the OP has anymore AP sound packs that he/she has installed as some are easy to check if they have installed properly for example, Class 303 - N button alerts the guard, Class 375/377 installs a single scenario in the L to B route and so on.
