Haven't played it yet, but my understanding is you just enable the "Be the Zombie" multiplayer mode in your menu. Those who then invade another player's game will be the night hunter, while the others will be humans.

Enable invasions in settings while playing DL and wait till night hunter connects to your lobby

you need to have invasions active and a night hunter will join your game if public but if its friends only your friend or u can hunt eachother
