The safest bet is on normal where you only lose 2-3k points during the day and nothing during the night. On hard, I believe it's 25% of your total(?), and on nightmare, well it's a nightmare scenario :) where you lose ALL your points, even at night.
I once lost 1.4 million points (was around level 50) just by drifting sideways with the grapple inside the stuffed turtle quarantine and falling flat on the electrified ground; didn't even get a chance to jump back :)
As BladeMaster said, the best way to level is to farm drops and deliver them in one shot to the quartermaster, in quantities sufficient to level up 1, 2 or x levels at once.
