I can't break 100 million score in a single hand, how do people even get to 40th ante or more?

Doing it with just jokers is hard, using steel and glass cards on top of whatever jokers you had is a lot easier. Try not to play your glass cards until you have a bunch from duping or using multiple justices, then go for one big hand at the end of a run. Duping a red seal glass card or red seal steel card is also obviously way stronger.
Most of the runs that go very far into endless mode rely almost entirely on steel cards and/or kings with Baron. But just to hit 100 mil only a few should suffice.

Unlock the Plasma Deck (14th).
Play the Plasma Deck on white stake and continue in endless mode.
Usually you start the game by boosting chip count, but if you survive the transition to boosting mult instead, you are almost guaranteed to hit 100M at one point. Plasma just yields ridiculous scores, that's its thing.

Or it could just randomly happen while not trying.
Yesterday, I had the luckiest run ever. I was playing the green deck on orange stake. I defeated the boss of ante 7 with a hand worth over 100 billion. Not a typo, not million. When you see the e11 appear, it's definitely not millions. I had DNA, Brainstorm, the Idol, Hologram and I don't remember the other(s). The idol had about 2/3 chance to hit the right card. At that point I was playing flush five hands of lucky red seal aces of heart. Total insanity.

There is a rare joker that can carry you way past that: Baron. (Kings held have X 1.5 Mult)

Getting it requires a bit of luck, which you can nudge with the rare joker tag, but the basic strategy is getting it and a deck with Steel modded Kings, then just play a single card and watch the multiplier climb.

Examples for useful jokers: Any money makers, so you can reroll for Baron aggresively,
DNA to copy more Steeled Kings into your deck, Brainstorm and Blueprint to copy Baron and Mime to trigger held Kings twice. I got to ante 18 with that one

This is absolutely the best strategy, regardless of deck ^^

Buy tarot packs for chariot cards to use on your kings, and death cards to duplicate them. If you get lucky enough to find deja vu in a spectral pack, use that to apply a red seal onto one of your steel kings and start duplicating that instead. Also hanged mans to thin the deck as much as possible...

The best way to do it is with glass and retrigger abilities (which is way more consistent than Baron, since that needs at 2 specific rare jokers to work, and another few specific uncommon/rare jokers to pop off). Assuming you play a Flush Five of 5 glass cards, 15 triggers of your glass will almost always be enough to get it. The best way to facilitate this is with any of the retrigger Jokers besides chad, a blueprint or brainstorm, and 5 of the glass you want to use, preferably either face cards or 2-5s for Hack and Buskin. You can also use red seals if you can't find a retrigger copy, or some other good X mult Jokers. You still want copy abilities, mainly Death and DNA, but it's much more reliable to pull off than Baron.
