So I reached over $400 in normal mode and didn't get any achievement...
The achievement says "in a single run", what does that mean?
Do I need to go over ante 8 to unlock it?

You need to hold $400 or more at the same time to unlock it and it doesn't matter whether you achieve that before or after going endless mode.

If you are using mods, there's also the possibility you got the ingame achievement and unlocked the joker from it, but not the achievement on Steam.

I can't believe it, I reached more than $400 in the shop just before the boss blind which I couldn't pass T_T

No, you just need to reach $400 or more. It does, however, need to be in an unseeded, non challenge run. So not during The Rich get Richer,

Oh so you mean just reaching more than $400 is enough, and that I can get the achievement directly from the store if I sell some cards?
Well maybe I was in a seeded run, that would explain why it didn't work T_T
