Am i required to buy a blank in order to get +1 joker slot one in the shop this run? Ive bought 10 of them already, but havent seen +1 appear at all.

Yes, all upgraded vouchers require the prerequisite voucher to be purchased before they will show up in a run :]

Yes, you need to buy Blank Voucher during a run before Antimatter will show up. All tier 2 vouchers require their tier 1 counterpart to be purchased before they can appear.

Just to be clear, the 10 times blank voucher purchase is just to unlock antimatter. After it is unlocked, it has a chance to appear in any run after you have purchased the blank voucher (in that given run). No need to buy 10 times anymore after unlocking.
