In my case, The Wall would be a heavy contender (extra large blind), The Eye if you never built secondary options or cannot win games in a single hand, The Needle (only play one hand), now that one can be a real nuisance without a strong build already going, you pretty much must win with your first and only hand....
From the final boss blind, oddly only two are a pain in my opinion, Violet Vessel to a degree, and last one of course my fave, most hated, despised, strongly detested, Crimson Heart. For those that haven't experienced Crimson Heart, i won't spoil the effect, you just gotta enjoy it during your first encounter in all its crap glory
The Eye can be scary if you're built into a narrow set of hands and can't one-shot it. (Like my latest run using mainly pairs with half joker and card sharp, though I pulled through.) The Arm less scary, more 'this is annoying and costly'. The Ox could break you if you can't score effectively with a secondary hand, though it's not necessarily fatal to eat the loss.
The Psychic hard-counters the half joker, which could be hard for some runs. Mostly harmless otherwise.
The Water has killed me when I had a run that needed a higher-end hand to score effectively, though it's not a problem for builds that use minor hands.
The Wall and The Needle are both usually not a big deal if you're doing okay, but The Wall gets harder if you've taken -hand effects.
The Plant is death on face builds, obviously.
Everything else I just file as 'annoyances'. The Pillar looks scary but usually isn't really. The Manacle might be a big problem if you've managed to accumulate a bunch of hand size reduction somehow.
You want me to clear the blind in one hand? Okay, let's see.. discard... discard... discard. Well I'm out of discards and the game still hasn't given me anything good enough to win with. Guess I lose... on ante 7.
Yes, this just happened to me.
If you couldn't dust the Needle in one hand with all your discards, you probably couldn't dust the Wall in four either.
(Unless you're relying on Card Sharp, obviously.)
Yes, but the Wall does offer a certain amount of leeway when it comes to bad luck. A bad hand can be balanced out by a particularly good hand.
With Needle, you can run out of discards without ever getting a good enough hand to win with. Then you're screwed.
If your scoring strategy is so picky 1+discards not getting to score is likely, but so powerful that it will easily crush the Wall if it does score, that seems like a bafflingly bad build choice.

Maybe it's just that loses against the Needle stick out in my mind more. I remember more of them. I've had several Straight runs lose to the Needle because the game steadfastly refused to give me a Straight.
I think the fish is a lot worse than needle/wall as Burgler can resolve these bosses relatively easily.
