Report a translation problem
If the hand you play is a straight flush, a flush house or a flush five, you won't get the achievement. It clearly says the hand you play has to be a flush and not "contains a flush". A 5 cards hand that contains a 4 of a kind won't work either as the game scores the 4oaK, not the flush.
In other words, you'll need wild cards of at least 3 different ranks to unlock the achievement. Use Strength tarot card to help you if needed.

I think one of them had been a wildcard flush five, but the other one was definitely not. Do challenges prevent achievements? I could have sworn I was in a challenge when I got the "discard a royal flush" joker unlock, but I guess that's not an achievement... which would explain why it didn't pop.

When you say you're playing a mode in which you unlocked some achievements earlier, it implies it's not in challenges.
Because you CAN'T unlock achievements in challenges. Except for 2 achievements that are tied to challenges.
I'm sorry what is a "challenge"? I am confused as to what that could possibly mean in the context of this game where there is only one thing called a "challenge". Your post is incomprehensible to me, what with all the confusion about what a "challenge" is.
In your OP, you said "during normal challenge and base difficulty runs during which I had also gotten other achievements".
"Normal", "base difficulty", "runs", and "during which I had also gotten other achievements" were all pointing to you doing regular runs, at white stake.
"challenge" is the only word that was not pointing in that direction. So as I said, I assumed you were not doing challenges. Turns out you were. But no biggie.

From the main menu, when you click "PLAY", you have options. "New Run", "Continue" (if you did not finish your last run) and "Challenges".
Now I know you were not doing regular runs. You were doing 1 of the 20 challenges.
In challenges, you can't unlock anything. No achievement, no new card. The ONLY exception is a couple of achievements that have to do with completing challenges.
So when you are hunting achievements, play regular runs, by clicking "New Run", don't click "Challenges". And once you're in a new run, my initial post describes what hands won't work for the achievement you're hunting.

No, malogoss said not to refer to regular non-challenge play as "a challenge," because that's confusing to everyone else. As you have indicated you were playing a challenge, so you only confused yourself.
You're the one that put "the only thing known as challenges in-game" into what malogoss wrote. I'll requote for emphasis:
Challenges, contrarily, are not "normal" nor "base difficulty" so do not use the word "normal" to describe a challenge.
When people are being both dumb and unjustifiably condescending online, I find it's best to goad them on and waste their time. I doubt it has a net positive outcome, but it's better than trying to engage with them earnestly.

Ironically, I used the term "normal challenges" in an attempt to avoid confusion and questions as to whether I was using specific seeds or mods or anything, but you can see how that has played out on this forum. People seem to be quick to be condescending and correcting here instead of asking clarifying questions, which you seem to only have done as an afterthought while attempting to rag on me.
In other words,
"others are dumb and yes, I'm a troll".
Whatever you gotta tell yourself man, gotta avoid self-reflection somehow I guess. You seem to have a pervasive character flaw of being aggressive and condescending given the interactions between you and other users I've seen on this forum.
