What you can't do is unlock anything or complete challenges, so the replay value of the game will be next to nothing. You won't even be able to unlock profesional modes for the 6 main missions that have them (which is a higher difficulty level)
Some of the things you can unlock are starting points, places you can choose to infiltrate during setup, instead of always being at the default location. Some of those locations include already having a disguise. They come from mastery of the levels, basically the game assumes you've played enough to know how to get to certain locations and disguises and just lets you pick that as a starting point for replays
You also unlock new smuggling locations, so you can place certain weapon/items at a pickup point of your choosing.
And then of course there's the variety of tools and weapon unlocks
Thing is, if you just want to complete the game once through, you don't need any of that. The game is 100% completable with the basic suit and no items at all (you technically dont even need the equipment you start with, every level can be completed starting with nothing if you wanted to).
You would still be able to pick up disguises, weapons and equipment during a level (and none of those are needed either, they are all optional)
You just won't have the ability to unlock things for future playthroughs, which will minimize your replay value
You also won't have access to player made contracts (challenges made by other players) of which there are 1000's and give the game near unlimited replay value beyond the 16 missions it comes with
So, can you play it offline? yea, but there's less than 10 hours of content that way. Played online with a goal of doing all the challenges you could easily get 50 to 100 hours out of it, and if you like contracts, that's as many 100's of hours as you are willing to put into it
