Use it to buff your estus flask. The women at mejula will give you additional stacks for your flask and you can burn some dust in the fire there, to increase the level of the flask.

If you are talking about using dust/shards after you have a 12 +5 estus flask, you can sell them to Gavlan for I think around 5600 souls a piece.

If not, then you'll want to give the shards to the Emerald Herald in Majula and she will expand your flask 1 charge, and burn the sublime bone dust in the bonfire in Majula to increase the strength of your Estus heal.
