The only difference is nobody will joing your game so you're on your own for the whole match.
Yes you can play the game offline without the internet connection as long as your steam can launch :p
edit: yeah idk if your perks stay the same, no idea if they are saved only in cloud
However: If you think of only playing offline, you miss out a LOT of fun. I'd say the whole fun comes from playing this game coop. Singleplayer - I would not buy the game for SP. The Perks complement each other and whole gameplay is tailored to coopplay.
