There are no profitable trades without a purser, and he should have at least two of the trade abilities.
If you buy a Dutch trade license at Curacao (the door with the gold sign on it) and can raise either a Spanish or Dutch flag, you can visit Spanish colonies without problems.
You can get price lists for other colonies by visiting merchant ships and talking to their captain, and sometimes you can get a price list from the storekeeper.
Hook
Another good town for selling goods - Capsterville on Saint Christopher. You can sell there coffee from Guadeloupe. This trade route very short but not such profitable as trade routes to Curacao.
I think that best prices for selling goods in this game in the local capitals (Willemstad, Capsterville, Port Royal and Havana). So maybe you can try some trade routes for last two towns. But I dont like their positions on the map.
And of course you need purser or minimum two unlocked trading abilities. I suggest you pick these trading abilities for indian officer Tichingitu if you have him. He always will be with you, so you can appoint him as a purser if your regular purser leaves you in tavern in a middle of trading route with goods in your hold.
It shows the most profitable trades from one city o another (please note you must be physically present in the city for this to work).
My favourite trick is to figh pirates, then take over their ships, then use them in my fleet for storage. Once I reach destination i sell it all.
I normaly use Xebec wich is pretty fast (17 knots). so when attacked I give order to the rest to flee. Normally all enemies will chase you anyways, so your fleet is safe.
I agree that Williamstead is best for trading.
The issue is that once you have a couple of millions there isn't much you can do with money. It is a shame there is no option to found your own tiny town and develop it.
Right now in my game Tichingitu has 3 personal abilities and 6 ship abilities that I can assign, so I guess I could make him anything. It seems a little gamey to use him for anything but a bodyguard, but that doesn't matter that much.

Hook
The Active Map is available as a stand-alone mod if people want to use it without using Hookmod and it works with any version of TEHO. Kb31 has basically implemented the map that is already in the game but unfinished, and I don't consider it a cheat. It's the best thing to ever happen to TEHO.
Active Map started off as a simple map that showed your ship's position. We added the island region boundaries and this makes sailing on the deck of your ship, going only to the world map to cross a region boundary, very easy indeed. It also had some information on other colonies, and I requested the trade assistant which will show you profits for each good. It also can give you the estimated travel time in several formats depending on whether you sail on the world map or on deck. I prefer showing the distance in nautical miles, and it adjusts for tactical or dynamic sailing mode.
The most profitable trade I ever saw in the game was rum from Trinidad to Curacao for 71 profit for each unit of rum. Santo Dominto on Hispaniola also has a thing for rum and if you're going that direction you can buy rum at Tortuga and a few other places for a good profit in Santo Domingo.
Hook
You can spend this money to maintain big fleet. When I tried to capture Tortuga without pirate help I gathered fleet of two 1-class and three 2-class ships with total crew near two thousands men. The month payment for this fleet was near 600 000-800 000 pesos. I even started to took back my money from banks. :)
