Secondly, maybe look at the Patch 17 Troubleshooting stickie. Might be something in there that could help.

The issue sounds vaguely familiar, so you might be able to find some posts by other people who have had this issue and found a fix. If not, then you can create a post about it. (Note the official support forums are community run, but are on the official forums so CA has a larger presence than here.)

I'll take a look at the others. But an initial google search for this issue yielded no results of any kind.
There was an issue with the launcher which I remember reading about that was similar, so maybe try searching for "launcher" rather than mod manager (if you haven't already.)
Just to check, is this the official mod manager/launcher, or is this a third party mod manager (e.g. Mitch's Mod Manager?) Mitch's one isn't meant to work anymore because it's not up to date.
It also could be that a mod isn't compatable with the latest version of Rome II and that's causing a conflict. Maybe try manually deleting all the mod files, then verify and then see if the game will launch properly with no mods.
Finally are you running Windows 10, in which case that could also be connected.

I've been using Mitch's for a long time now, since it is better at letting me control my mods. I never had problems until this latest patch.
Thankfully, I'm not using Windows 10. I'm still running 8.
