Well, it took roughly 8 years after the final content release for Napoleon to get a Definitive Edition. So maybe sometime around 2026 for the Rome 2 one.
I definitely wouldn't expect it any time soon.

Emperor Edition - A quick guide.
Emperor Edition contains the base game and all the free content, specifically:
•Rome II Base Game (8 playable factions, Rome, Carthage, Macedon, Iceni, Arverni, Suebi, Parthia, Egypt.)
•Patches 1 - 15 (including the Patch 15/Emperor Edition major overhaul.)
•Imperator Augustus Free Campaign (10 playable factions in that.)
•6 Free DLC factions ( Pontus, Seleucid, Baktria, Armenia, Getae, Massilia)
•Free Battle of Pydna Historical Battle.
You'll also get Patches 16, 17, 18/Power & Politics, 19 and 20/Ancestral Update automatically downloaded when you install.
(Note: Mac users will only be updated to Patch 17, as 18, 19 and 20 haven't been released on Mac yet.)
Emperor Edition does not contain the paid additional content, specifically:
•5 Culture Packs (Greek States, Nomadic Tribes, Pirates & Raiders, Black Seas Colonies, Desert Kingdoms)
•5 Campaign Packs (Caesar in Gaul, Hannibal at the Gates, Wrath of Sparta, Empire Divided, Rise of the Republic)
•2 Unit Packs (Beasts of War, Daughters of Mars)
•Blood & Gore DLC.
(Note: Mac users can't use Desert Kingdoms, Empire Divided or Rise of the Republic because Mac users are still on Patch 17.)
Emperor Edition is a rerelease that accompanied the Patch 15 Major Overhaul, rather than a Game of the Year edition. (Also it's a free update for anyone who had Rome II, or whoever buys an old disc copy of Rome II even now. It functionally replaces Rome II, so everyone who plays Rome II has Emperor Edition these days.)

If you mean an edition with all the paid-DLC included for the base game price, there isn't one, nor in my view is there likely to be one for many years to come.
There's no real need for one. The base game has a huge amount of content; the DLC is completely optional (you only need it to play as those factions, not to fight against them in Grand Campaign); Rome 2 continues to be in the top 3 most played Total War games and top 100 games being played on Steam; and it still apparently sells well, as there's a noticeable influx of new players especially during sales.
Also the number of players waiting for an all in one edition appear to be a very small percentage of the community, and even if they were to release such an edition it would have to cost a lot more than your typical definitive edition to be anything like representative of the content included. Which comes with its own drawbacks, as people would inevitably complain at the price for an "old" game, even one with 36 playable factions in Grand Campaign, and 6 other campaigns which brings total playable factions to 70+.
There's basically no "up side" for Sega (the publishers) nor CA (the developers.)
What we may see is more editions like Caesar Edition (Rome 2 + Caesar in Gaul DLC,) Spartan Edition (Rome 2 + Greek States DLC + Wrath of Sparta DLC + Daughters of Mars DLC,) and the most recent Enemy at the Gates Edition (Rome 2 + Hannibal at the Gates DLC + Empire Divided DLC,) bundle's of the game and specific DLC released in physical (and occasionally digital) form in certain countries.
I know, probably not the answer you were hoping for. I just wanted to give you the full picture of why I feel a "definitive" edition is unlikely in the short to mid-term. We may get one eventually, but for now you're probably better off looking out for sales and grabbing the game and what DLC you are specifically interested in in one of those.
