As I understand it:
Culture Packs, you will need them if you want to play the DEI versions of those factions in Grand Campaign. The same is true for the factions added to the Grand Campaign by Caesar in Gaul and Hannibal at the Gates.
Campaign Packs, you only need them if you want to play the DEI versions of those campaigns. (Some have been more DEIfied than others. Wrath of Sparta I think they changed it into a different conflict entirely, while I don't think they've done much at all with Rise of the Republic yet.)
Unit Packs, no idea.
Blood & Gore, only if you want Blood & Gore.
I think that's right, but as I don't play DEI I cannot say for 100% sure.
