It will work fine with win 10.
In a few words:
1- You must add one dll. file inside the correct game folder.
2- Add a simple parameter at the game "launch options" through your steam library, (game parameters)
3- Activate Mother VR inside the game, exit the game
4- Activate Steam VR
5- Enjoy.
That basically activates files from the original game that makes possible to play the game in VR. Because this game was originally inteded to be used with VR but at the end this function was desactivated by the developers, but some files where not delated and now is possible to reactivate all with that patch.
About the controller... I have Index controllers and you can make the whole thing with them... but is a pain in the ass because you can't do some interactions with some things inside the game. That force you to move up and sit down all the time to use controllers for the most part and in those cases where you can't use your controllers sit down to use the keyboard.
My advice is to play the whole thing with Keyboard and mouse and enjoy the VR views.
With practice you don't need remove your headset to see the keyboard at all.

Do you have windows 11? Did you activated mother VR inside the game through it's options, exited, and entered the game through steam vr?
edit(because im nice):
Guide to fix:
Go to Steam\SteamApps\common move (cut & paste) the Alien Isolation folder to somewhere outside of Steam.
I went with D:\Games as that's the same drive I have Steam on.
Rename the Alien Isolation folder to AlienIsolation.
Delete the Steam\SteamApps\common\Alien Isolation folder if it still exists
Open a command prompt and enter mklink /d "Path To Steam Alien Isolation Folder" "Path To New Location"
e.g. mklink /d "D:\Steam\SteamApps\common\Alien Isolation" "D:\Games\AlienIsolation"
