They're not mandatory though.
You wouldnt spoil anything either if you choose to watch the films later.
If you decide to play the game first, you won't know anything about the setting and the elements of Alien and you will have the sort of 'Realistic-immersive' kind of experience, because in this case the player is just as clueless as the main character he/she is controlling. It can be very immersive but it won't have the 'Already-forged' bonds it would have if you have watched the movie first.
If you decide to watch the first movie first, then you'd be acquainted with the setting and the background of the story: that would provide you much more interest towards the characters, because you already have the tools to decipher some of the Alien universe's language and give you that "Aww! I know this from the movie!" sort of feeling. Although, that might ruin quite a few moments in the game that if you haven't watched the movie first you'd be totally blown-off by.
They are both valid choices if you ask, as there are pros and cons in both. It also depends about how you want to be scared. It is the anxiety of knowing what's going down on you that gives you the higher suspence and thrill or it is walking into a complete unknown territory?
In any case, Isolation is a wonderful game and you would be pleased either way. I have written a review when I first played it, check it out if you had not read any yet.
So watch first movie, play this game, watch the rest.
...skip Prometheus. Good concept, ruined by the terrible writing of Damon Lindelof (the guy who ruined LOST). The only way it could've been worse is if M. Night Shyamalan wrote it.
