Yeah the easiest one to miss would be Clyde himself, because you can only friend him right at the beginning of the game and its a one chance only time, you miss him you miss out of the "get all friends" achievement.

Also one thing, either avoid Al gore's entire side quest until you finish the game or put up with his spamming because if you "unfriend him" aka going to fight him because apparently that's how someone unfriends someone from Facebook in south park that also makes you miss out of that achievement.
