If this were me, these are the things I would check:
Firewall. Ensure that your firewall is not blocking your connection.
Port Conflicts. There are games that can use the same ports or port ranges as another. As far as I know, SR4 doesn't need port forwarding, so make sure that all your ports are off.
Check more than one person. If you are only attempting to connect to one person, try to connect to someone else. It is possible that the problem is on their end and not yours.
Only 1 of my friends bothered to get the game, more precisely my gf.
Another friend is picking it up next week, but that's not very optimal wait time to find out if the issue is on her end or not >.>;
I have also faced the same problem.
What wonder why it cause?
Already been tested, the only thing I can tell that could cause issues, is ports, but I can't find any port information for this game.
What fixed it for my brother and myself was the typical Steam fix:
Go under properties of the game (in the steam client), and tell it to verify the local cache files.
We also made sure that the option to automatically keep up to date was checked, but the verify cache was what fixed it...
another friend just installed this game tonight and I am now having the same issue connecting to him.

Since the game files themselves haven't changed that would indicated that something has changed on your end with either your hardware or software. Unfortunately, that can be just about anything; additional software like hamachi, windows updates, changes to Windows firewall, etc.
You mentioned rebooting your PC. Have you also tried rebooting your router and/or modem as well?
For others having this problem, SRIV uses the standard Steam matchmaking ports. Do other Steam games work correctly like TF2, Left 4 Dead, etc? Here's a list of standard ports:
