No one has been able to explain how opening this door would cause the bug, it has just been observed that the bug occurs after opening it, and replaying from before that point results in the bug not occurring. It may have absolutely nothing to do with the door, but loading a save before entering Holger square and not touching the door has the best chance of getting you around the bug.
No, it's a bug. Every time you go through the level they should enter casually (even if a nearby alarm was rung). The bug is caused by opening the gate after you free Overseer Martin. All it takes is a simple level restart.
Edit: I'll make a video showing that it happens when you open the gate and post it here.

Stupid bug nonetheless - it almost made me not buy the pretty neat game...
It is the iron door after you free Martin the Overseer. At the place where the game asks you to blink to cross over obstacles. Once you are over, you should be able to find a lever that opens a door besides it. Flipping the lever causes the bug to happen. The player wasn't suppose to flip the lever, Overseer Martin should.
At the meeting room (Without the bug). If you switch the two glasses. Campbell will be poisoned and the other overseers will come. After a quick quarrel, Martin and his guards will kill other overseers and try to find a way to escape the facility. This is when Martin runs all the way back to the iron door and pulls the lever.
I believe the game detects whether the lever is pulled or not to trigger the overseers aggro towards Campbell. Which also explains why Campbell, Martin and the guards ignore the player after they rush out of the room. Campbell is not looking for the player, but Martin and his Guards. And Martin is also not looking for the player, but to escape. My guess, not 100% sure :/
