The dunwall city trials is just a challenge mode type thing. It gives you missions to do seperate from the stories that you can do anytime.
The knife of dunwall and brigmore witches are really good story dlc's and should be played after the main story. Knife of dunwall should be played first :)
On the other hand, The Knife of Dunwall and The Brigmore Witches are two parts of a separate story that takes place in Dunwall. Without giving too much away, you play as Daud, not Corvo, and it takes place at the same time as Dishonored. You get to revisit Coldridge Prison and Daud's Base, but the rest of the story takes place in new sections of Dunwall, and the countryside around. Knife of Dunwall is the first, and Brigmore Witches are the second.
The feel of the game is true to Dishonored, though I found the DLC to be much more difficult. One of the perks is that you control and weild some new powers from the outsider, and you can find a lot of lore. It is definately worth the purchase.
As for the Void Walkers Arsenal, (which includes the Arcane Assassin, Shadow Rat, Backstreet Butcher, and Acrobatic Killer packs) it's not really worth it, except for the feel of owning the entire game. They add some money and books, as well as a few bonecharms. If you're into that, go for it; just know that it doesn't terrifically effect the gameplay.

*edit* I totally misunderstood the original question: you can play the DLC before, during and after, and it doesn't really make a difference. The end of the DLC story coincides with the end of "The Flooded District" chapter.
As mentioned, powers and weapons are a little different in the dlc's. Many players seem to like the improvements which you may miss in the base game, if you've played the dlc's first.
As for the Dunwall City Trials, imo, they're exercises in trial and error. Simply a series of challenges unrelated to the story of the game. For some players, they are fun and rewarding. For others they are nothing but a source of frustration. It all depends on your play style.
