Note, though, that not all titles are playable. You can't play a bishop (such as Hamburg in the 1066 starts) a non-merchant republic (such as Ancona in the 1066 starts or Genoa in the earlier starts). Additionally, there are a lot of titles in the game that require DLC to play-anything that isn't a christian count, duke, king, or emperor is either unplayable or requires certain DLC to play.
You may also have corrupt files, try using the verify local files action in Steam.
Also what specific message pops up when you hover over the play button?
Roughly translated, it says "not allowed to play"
There should be a reason listed such as "Jewish characters require Sons of Abraham DLC" or "not allowed to play theocracies" (assuming the localization includes all the stuff it should if not playing the English version).
(with the exception that you can't start a game with any character past 1335 or so, but i presume you know how to change the start date etc.)
Quit Steam, delete the game files folder for CK2 in Steamapps/common,
start Steam again and reinstall.
