Maybe one can't even trigger now because the only den leader is gone and has story relevance? So the game never attacks his den becase he needs to be able to get his position back? Idunno.
If you actually enjoyed it and want to do more of them, there's a sequence later on where notoriety is forced on you and doesn't go down I think. However since all your dens are already defended by masters, you might need to reload an earlier mission where at least one other den doesn't have a master assassin in it and try again with the notoriety.
so I get credit for completing the Den Defenses (for the challenge) even if I don't actually do them myself?
Once your notorioty reaches max you still have to perform a few high-profile actions. For me when trying to force a Den Defence I get notorious and then just go rouind performing high-profile assassinations on guards. I think I usually have to attack 4 groups of guards before the Den Defence is finally triggered.
But I have noticed that occasionally the you cross paths with the nobleman guy - can't remember his propper name, but it's the guy who has 2 guards following him - and you automatically get into a fight with them. The problem is, I think killing the nobleman instantly lowers the notorioty status and puts you back to secret status, so you need to run away from the nobleman if you see him coming.
I also think you can just replay the memory with the first Den Defense and victory will count towards the guild progress.
the noblemen guys seem to only drop notoriety by half, so it is still possible to get back up, and one the 3rd den defense that I let trigger it actually happened with red flashing, and like 40%.
