People who say it must be transformed are actively wrong. You just need to be ABLE to transform.
I think the achievement requires you to play on Castlevania map, transform Cornell as soon as possible, and then do a certain number of kills with him transformed.
I tried that by killing about 100000 mobs and unlocked him. But I can't affirm that's the number, since I retried the run other times with Cornell transformed.
Transforming as Cornell as soon as possible only contributes to obtaining the kills quicker, and is not necessary to begin the count of 100,000. Like every other Megalo secret, you only need 100,000 kills in total.

Not for Cornell somehow. I did 3 runs as him on Castlevania Map, and didn't unlocked his megalo form.

You probably are missing what I was missing: the Pile of Secrets.
It's a relic dropped by the boss on the north west, the hanging zombie
Once unlocked, you get an extra list of characters and the doggo is among them
I had Pile of Secrets already unlocked; I was making my way down the list and had just unlocked the secret characters above him.
Tried again in Ode to Castlevania, this time did not pick Ebony Diabologue and amassed around 140k kills. Got it this time so I'm thinking it might have been because I didn't select one of the default weapons. Either that or the "slay the Castle's denizens" bit meant I had to be in the Ode to Castlevania map. I'm not sure which it was in my case!

The hints are very precise: you have to evolve the weapon (turn into a beast) AND do it in the Ode to Castlevania (slay the denizens) map
So it seems the spawned items from selection boxes is absolutely not eligible to contribute to anything even if logic says they should.
