It gets larger the longer you're on the map, and eventually it's primary hitzone is off-screen and it has more health than a death/reaper does, at which point it's basically unkillable.
But you can very, very easily kill it within the first minute or 2 with any remotely powerful character.
Personally did it with death, but leda probably makes more sense. Alternatively, any character you have massively egged.
Get a few levels for weapons and then immediately run to the question mark to fight the ball. Pick weapons that target the nearest enemy and weapons that can be directed. Dagger is a good pick. Make sure you get in front of the katamari so that your knockback will keep it from rolling forward, this will slow how many enemies it absorbs and keep it on screen so that you can press damage. If you wait too long, it'll be too strong to reasonably kill and you'll need to try again.
If you've unlocked the Yellow Sign, you can instead go for Infinite Corridor to cut its HP in half over and over until it's killable.
Then take Leda into Bone Zone and immediately head south. Catch the Ball of Bones on it's trek right and kill it with the evolved magic wand.
It doesn't have to be Leda. You can use Red Death or Marrabio too.
I don't consider this the best game design no. If it were a secret, I'd be okay with it. But it's a collection item and on the map so it shouldn't get to the point where it's impossible to kill unless you look at a guide or someone tells you.
I do find the boss fun though and had a blast when I didn't figure it out the first run and it was massive.
30 hours is rookie numbers.
Red Death very easily kills the thing, and honestly, I'd hardly call them a secret character. You will naturally unlock them if you just experiment with the new things you unlock as you progress. As I said in another post complaining about this, all you need to do is take Red Death, take Silent Old Sanctuary, and beeline straight for the bone katamari. Once you're there, stay in front of it and keep as close as possible to maximize your damage. The knockback from you weapon will keep it almost stationary. Doable even with eggs turned off, and done in less than 2 minutes.
Died in 2 minutes and unlock was confirmed.
You can wait until you have a late game build to fight it, since it seems to spawn at the ? on your map everytime, just don't approach it until you're ready
Death does not seem to spawn if you are fighting the bone pile until after the bone pile dies.
After you kill the bone pile your first death will spawn after the next one minute mark so you get to explore an empty map mark for up to a minute depending to on when the bone pile dies.
EDIT: Taking this approach meant I did 1G damage with Infinite Corridor so no idea how much health I ended up giving the bone pile
