Go to C:\Program Files (x86)\Steam\SteamApps\common\Mass Effect\Binaries
Right click MassEffect.exe (if you don't have extensions showing it will just be MassEffect without the .exe)
Click Properties
Click the compatibility tab
At the bottom tick "run this program as an administrator"
Click Apply/Ok
Then try and run the game again.

I dont need to uninstall and re-install the game. After browsing through the files, I found the Launch.xml file. Since its there, I dont know why its not working.
Im using an Nvidia graphics card. ive forgotten the spec's but the card is fairly new and costed me around 300$.
Ive tried to initalize it an it still wont work.
I HAVE TRIED RUNNING BOTH STEAM AND MASS EFFECT BOTH AS AN ADMINISTRATOR AND IT DIDNT WORK. With that said, I need to try something else...
1. Rather than try to launch Mass Effect from Steam's desktop icon, go into the game folder and make shortcuts (copy the executable and "paste shortcut" on the desktop). MassEffect.exe is in the \binaries folder inside the game folder. Set its properties to "run as administrator". You can do the same for MassEffectLauncher.exe in the main game folder, but it's probably not necessary except for the initial configuration of the game on your system and the "Play" button won't work anyway. You should right-click the desktop icon and select "run as administrator" to launch.
2. Go into the game's \binaries folder, copy PhysXCore.dll and paste it into your Nvidia folder (Program Files (x86)\NVIDIA Corporation\PhysX\Engine\v2.7.2\). You will likely need to create the \v2.72 folder and this is a useless idea for Radeon users, but maybe they aren't having this issue.
Tried that, but Origin wouldn't take my Steam CD key. The "Pinnacle Station" DLC that I got from Origin downloaded and installed without a problem, even though I ran the game through Steam, so I avoided dealing with that headache. I did need to redownload and install "Bring Down the Sky". Bioware made it available freebies and the link is at:

I bought an Ecopy of it on steam. Not to mention that what you're sugesting sounds like a serious headache. I have all the files I need to run the game, its just a software issue though and thats what we need to focus on..
2) right click on MassEffect.exe located in Mass Effect's Binaries folder(steam/steamapps/common/Mass Effect/Binaries).
3) select properties (scroll down to bottom)
4) select compatibility tab
5) in compatibility mode, check "Run this program in.. for:" check box and select Windows XP SP3
6) Save this change
7) Run ME from me.exe. Steam should open and ME should run. You can open ME normally (within steam) too.
This process worked for me. Hope this helps
EDIT: In fact, I went and looked for the binkw32.dll file and its right there over the ME.exe file.
I installed this game today and had the same problem, I think I fixed it by executing the <<runme>> aplication (in SteamApps/common/Mass Effect folder) as administrator. After that, the game is running fine through Steam.
