If not try verifying the integrity of the game
First, I suggest you install the 1.02 Patch[masseffect.bioware.com] for the game. In theory, it should already come with the Steam version, but some people managed to solve their problems by applying it.
Second, make sure the game's .exe is running on Admin mode. Nobody knows why that happens, but it seems to also fix some issues. Compatibility mode (XP) can also help.
If both fail, consider activing the game on Origin, and then re-downloading it through there. The Origin version also comes bundled with both DLC. You can download BDTS and PS if you stick with the Steam version for free also, but I no longer have the link for that.
After doing this, you should be able to at least get in-game. Unfortunately, as I said earlier, I never really got rid of the annoying GPF error, which would crash my game often during loading screens. I managed to finish the game just fine but, if the same happens to you, save often; there's no definite fix that I know of.

Select Properties
Click the Local Files Tab
Click the Verify Integrity of Cache button
