Deluxe has more content in it (but not all of it). Standart version is missing a couple of missions and some other stuff.

Can you buy the extra missions later on if I don't get the deluxe?

Yes absolutely.
It's named Deluxe Pack (instead of Deluxe Edition) and adds three DLCs to the standard package. You can also buy these DLC individually.
