Nothing spawns back if you clear out the world. But there are root seeds you can plant to grow more roots for wood. As for enemies you can set up spawning areas with the spawn tiles you find on the ground. As for blocks - if you mine out the entire world, you'd have to start a new world to mine more. And of course you can plant seeds for crops.
Works especially well with the new world generation settings.
