i actually havnt played through since the ocean update so ill have 3 new biomes to get through
The update is only 2 days away. I'd say buy it now, get your bearings with it for a couple of days, then get excited for the update! :D
For a blind playthrough you're probably looking at around 50-100 hours to kill all unique bosses depending on your skill as a player.
You could honestly start your playthrough now or after the update.
While the major update that's coming in 2 days (10/4) will add new stuff to existing biomes the game is made in such a way that it generates the world as you explore so you should be able to find whatever gets added to the early biomes even if you start today.
That said the update is also just two days away. If you have something else you're currently playing I'd just keep going with that and then start your playthrough after the patch. That will also make finding new stuff in early biomes a little easier since the entire world is unexplored.
There's no wrong choice in this situation. It all comes down to personal preference.
One thing you'll probably wonder about is World Difficulty so I'll explain it real quick. Hard difficulty does 3 things, gives enemies more HP and damage (somewhere around 2x more of both, I think) and makes bosses drop more items per kill. Hard difficulty currently does not have any unique drops though.

It's far from being a novelty of the genre, but it does what it should do and does it well. There's still a couple of kinks with it, but is meant and works as a truly great multiplayer experience, without being obnoxious.
I'll maybe detail all that tomorrow, but I'm going to sleep for now.

Demo worlds, as long as you still have the local or steam save, are still playable. :) I've had my main world since the demo and it still runs just fine. :)
