Also happens to me a lot, the microphone could drop out randomly at anytime and cannot be fixed with any in-game settings adjustment unless the game is restarted. This is really annoying.
Also, testing the microphone will 100% make the microphone stop working.
When this happens, windows shows game is no longer using the microphone device.

There are many potential causes for microphone issues, but ultimately you will have to troubleshoot potential problems yourself. In the sound section, you should see the microphone you use in the Device section.

The game automatically detects what Windows is using as a default microphone, and if you do not see your microphone here, you need to change your Windows settings such that it’s the default.

If the game is set at Voice Activity, and doesn’t seem to be picking up your voice, try changing the orientation of the Echo Cancellation setting. If it’s off, try turning it on, and vice versa. Echo cancellation is usually helpful with older microphones, but may be detrimental in newer microphones that have their own form of echo cancellation.

Lastly, if you are using any software or applications that already use voice channels, such as discord, please close them first as they may interfere.
