If you're not max seeded with every unit unlocked then I recommend doing ending 1 a couple of times so you can progress further and get stronger creatures.
Btw, I am at about 5000 hours played currently. - Games a slow burn. You can try to get some inspirations or sins as well and get some of your base stats up.
You *CAN* brute force it with gluttony, but if you're to the point to where gluttony is going to take a month to show any progress, you'll probably have more fun just reincarnating again and working up those creatures and inspirations.
Of my many hours played, I only long-term gluttony grinded a few times, Once to kill the desert boss, and once to kill the story ending 5 boss.
iirc, it was about 5 ed1s to get ed2, 100ish eds1 to get ed4, and about 300 for ed6. might take a few more as many of these were long runs before greed was added to the game.
Number of clears needed is most likely dependent on sins/run though. Exp makes a big difference.
I don't think it's a lack of destiny that's stopping me from progressing, ED2 is Moment of Peace & i assume I have to beat the Demon King in the 2nd dungeon, but right now he's 2 billion HP to my 40 million HP & I don't see how to bridge that gap.
Excluding the (honestly insignificant) sacred ritual from ED3, endings 1 - 3 give you only more destiny. Selected ending has no effect on sins (the thing you are after now).
Afaik you should have unlocked Destroyer - make sure you get all summons from it (killing it after reincarnation without gluttony and upgrade actions will be harder than before reincarnation). 40M HP means more than enough stats to kill it. Seeding them isn't a top priority, they will take ages like all future guardian summons.
More greeds are unlocked with more reincarnations. They boost different things and make raising talent easier.
When I think about it now, many summons probably use their default skill because everything using set skills isn't always sustainable. This favors MAtk - more elements/energy pools to choose from.
