If you just look at the map and proceed to the next red destination without reading The Warren Report, you will miss out. In some cases, simply reading the item under Talk will create a new red destination on the map. Other times, you have to surmise from the Talk item and go back to a (non-red) location.
Plenty of extra dungeons, many super bosses (Dark Knights / Heavenly Generals / Overpowered Palace of the Dead units), and as noted above, you can go back in time to play through other story branches you didn't take, saving characters that may have met a tragic fate the first time around.
There's so much endgame content that the game is honestly a 40-60 split between main plot and optional.
