If you are referring to an ongoing game then just send all but one out to die in a bug nest or run butt nekkid into an attack wave.
I do solo runs all the time, but I recommend using a mod or two to even out the difficulty because you will be much slower to get those turrets going. Attack wave size modifier is helpful. I've also been trying out Jaggid's moons and running with Io to reduce frequency of attacks. SAD Comon Lib has bug fixes, and Terrain Recovery keeps you from deforesting the place. Jaggid's Survivor Tweaks pairs with Gender Equality so that women survivors don't move slower than the men.
Fast Feet is also a useful rule to use, both for general speed and to help kite bug attacks to thin them out before the real attack begins.

When you at some point want to see your defenses tested by a small or larger attack you just move the slider up.

you can set negative starting survivors to reduce the amount you start with
