I would differ from ShockedHearts and say that the story differences themselves aren't that substantial. The intermission, while fun to play, really just bridges the existing gap in the story.
Tickington/2D mode also offer a lot content-wise but I find it hard to consider it part of the game's "canon" myself. It's really just taking you through segments of previous games of the series. Some superbosses[sites.google.com] at the end, Act 4[www.nexusmods.com], etc. All are a blast, but not part of DQ11's story imo.
That said, I'd say everything else (bonus content / QoL / etc) culminates in a really satisfying experience for a returning (OG) player. You won't really be "missing out" by skipping this one, but if you're tempted to give DQ11 another playthrough DE is a good way to get a lot more mileage.
The ability to skip cutscenes alone is a godsend if you're tired of hearing the same lines over and over. (That and/or JP voiceovers)
