Afaik, it does not affect any questlines, but who can be 100% sure...
Not the talisman itself, but rather teleporting to the Divine Bridge and then having to sit at the site of grace there.
I used that shortcut numerous times. From what I remember, it does not count as an 'Altus Plateau' visit. Also, when checking the summoning pools list, that has been added as a feature when the DLC came out, the Leyndell area is not part of the Altus Plateau.
If you don't mind spoilers:
If you manage to activate the lift at the Divine Bridge without entering Leyndell in the 'traditional' way, it means you can access Leyndell without 2 Great Runes, which is the normal 'prerequisite'. (Only an invader would be able to activate the lift if you got to it from the Tower of Return)
It doesn't move Rya, nor does it advance any other quests. It won't get you to the Roundtable Hold either because it is not an open world grace.
