Some, like the Spiked Caestus, inflict bleed naturally, so you've got both blunt and bleeding damage in one weapon that hits fast (especially if you're two-handing it), meaning rapid buildup on susceptible enemies--and solid striking damage on those that are not (like the Valiant Gargoyles).
You're going to want high VIG, too, since you're going to have to get right in the enemy's face to do damage (fist weapon range is minimal).
Beyond that, I tend to stack at least 30 END; punching doesn't have a huge endurance load, but the ability to stay in the scrap will serve you well in both PvE and PvP.
(Note: Some fist weapons--like claws--benefit from quality scaling, so you'll want to balance STR and DEX; look at the weapon you want before really locking in on a forward-facing stat.)
Source: Completed base game and DLC with a fist weapon build (Dark 2 as well, but that game was broken for STR builds, making the caestus a straight-up murder glove).
u can tie that up with milicent's prosthesis and rotten winged sword talisman, alexander's shard, maybe add godskin swadling cloth talisman for health recovery if u get hit while punching :p
and leda's armor gives boost to attacks after a roll so... u can roll and punch perhaps ? XD

Dryleaf Arts. It looks badass but, at least for my part, I found it underwhelming and less effective than the beast claws I picked up right at the start of the DLC.

The Dry Leaf Arts punch weapon is very under-powered as detailed in this video that highlights a build based on the new Dryleaf Whirlwind Ash of War.
