When your seal is in your hand you use the light attack L1 or R1 to use the incantation or spell. Also the spell and incantation has to be highlighted and lit instead of being greyed out, so you'll know you have the stats to use the effect itself.
And yeah, don't two hand your weapon if you want to use a seal.
Make sure your weapon that you're trying to buff doesn't have any effects on it, like bleed or whatever.
