Can you 100% achievement this game in one play through

What the title says assuming I get all three endings with back ups, are there any quests that disappear if I do other quests etc.

you would need to get all endings and make sure you make a manual save before the last boss then re do save until you get all endings but i think it takes 2 runs

Yes, you only have to be careful not to miss the few missable achievements during the playthrough and then have to keep a copy of the savegame before finishing the game. Apart from that you have to complete the quests that unlock the different endings and that can't be done without spoilers as you need to know about them before, either by reading up on them or asking someone that played those quests.

There are 3 endings in game (for achievements wise). It is up to you - to make 3 runs or to make save backups right after final boss (steam cloud OFF). One natural ending you will receive anyway, another one - Ranni's questline (Witch-doll with big hat) mandatory for some other achievements and ending. Last one - Frenzy flame ending, you can progress up to ugly door (just don't open it). After final boss fight you can chose one of those endings. After final boss fight (and after ending, if you pick NOT to start new game) you can still make other achievements, BUT:
- you must not kill black fast beast (Maliketh in Farum Azula) before obtaining Bolt of Gransax (Huge spear in the Capital). It is better to find all legendary weapons before fight. And same you can do for Legendary Sorceries and Incantations. Only few talismans and ashes you can find in new locations, after Maliketh death.
You can beat most of the bosses too (actually, all of them).
- you must not kill Fia (women, that likes to hug people at your base (round table)) and you need to bring her Cursemark of Death in order to receive access to beat Lichdragon Fortissax for achievement.

You need 3 of the games 6 endings, which be of which locks you out of the other three. It can be done if you make a manual save prior to beating the main boss.

Do the summon ranni and the mend the elden ring options first after reloading the save each time, then go to the subterranean shunning grounds, kill mohg the omen, find the secret within his boss room, and get to the three fingers. Strip down and get the frenzied flame, kill the boss and get the frenzied flame ending.

Honestly would not reccomend doing this you are REALLLY missing out if you skip new game +.

If you do saves before the final boss you can do it in one run. However, there is one item you can lock yourself out of on the first run if you do something else first. If you do you will have to replay half the game to get it.

Ill be doing this in new game plus I just didn't want to do a third run if I missed any achievements
