How does lone wolf work?
It says single play but what does that mean?

How does it work with multiplayer?

You can only play "Lone Wolf" as a solo, but you can scale the enemies like if you had 1 to 3 other players with you, hence the "Lone Wolf" difficulty, the only difference from multiplayer is that you're all alone against up to 4 player difficulty settings.

Also seems like the enemy damage isn't scaled up.

Only works in single player

I hope they make it allow in multiplayer though

Why would they include it in multiplayer? The whole purpose of the mode is so that you can play singleplayer as if it was multiplayer. Although the enemies are lacking the attack and HP modifiers, the main reason they give you an extra blessing is because of the extra enemies you have to fight by yourself.

It would make sense to have a version of it that can make a duo or trio play as if there are 4 players. Since players would have access to the revive mechanic in multiplayer you wouldn't need the extra blessing.
