-Stuff You Keep:
Unlocked new weapons.
Unlocked new scrolls.
Spirit Essence
By "unlocked new scrolls/weapons" I mean that every time you fulfill the requirement to unlock a new scroll/weapon, the scroll/weapon will drop in the game for you to try out. After than the scroll/weapon will be added to the random drop pool for future runs.
As mentioned by Toxi, Spirit Essence is the currency used to unlock permanent upgrades under the Talents menu. There's general purpose Talents that will be active no matter what character you're using, but each character also has a handful of character-specific Talents that buff their particular playstyle.
The only Save function is a "Save and Quit." You have the opportunity to do this in the small room after every boss fight with a healing well/campfire in it. Click the well/campfire then quit the game. You'll see a Continue option on the main menu where you can pick up from where you left off. One thing to note, however: this doesn't work in multiplayer. Also if you have a saved run and you go to play multiplayer it will delete the save (you're warned before doing it and have to confirm the deletion).
