Yes with Moonshiner you must set a location for the Shack.
Only then will Maggie and the Frenchman be available for Moonshiner Missions, as they are inside the Shack. Also you will have random open world stuff that pops up at random that might be Moonshiner-role related; such as...

> Take out the Roadblock
> Rescue our buyer, who has been kidnapped

Also inside the Shack, Maggie has a whole lineup of Moonshiner Story related missions for you to do, one at a time.

Make sure to also double check your Satchel from time to time for "Recipes"
In RDO, if they do not have a checkmark next to them in your list, those are unread. Make sure you click READ in order to learn them. As over time you will find and learn other Moonshine recipes that can earn you more $ per batch.
