It depends, do you have the achievements "Give to the Poor", "Breaking and Entering", "Errand Boy", "Friends With Benefits" and "Lending a Hand"? If the answer is yes, you don't have to repeat the story. If the answer is no, I recommend that you search the internet for the achievements that you are missing because there are some that can be completed in chapter 6 yet (though only up to a certain point of chapter 6). And if you don't have any, you must restart the main story.

Just like the commenter above, it really depends on what you've already done throughout the game. Some mission strands are not available from certain chapters on (cannot say which from the top of my head), which then makes it unable for you to finish them. Not all are needed for 100%. Therefore, I suggest checking your own progress and checking online checklists of 100% for RDR2. If you need to do loads of collectibles and you're already in chapter 6, you may get bored if you cannot mix them up with normal story missions. The latter is the reason why I've started a new save to get 100%. It allows me to replay the game completely after about a year of not playing it, as well as obtain 100% without getting bored by merely grinding collectibles.

Another argument for saying that a second playthrough may be best for a 100% run, is that you've then already finished the game. You've finished the story, so you won't get any spoilers in case you need help with obtaining the 100% by checking maps for collectibles. That's also what I did. I played the game just like I would do, finished it with having done most of the stuff. I actually finished the game 92% in my first playthrough. Then after a break of a year I decided to get back and give the 100% run a try. That way, I also did not receive major spoilers regarding the world in my first playthrough.

Hope it helps!
