as far as single player. i see in a google search that a blue dot can mean ally. but it does not say whether this is also refering to the online portion or if its referring to single player story mode.

it shows up any time you are near someone who you are on the same side of, like when uncle comes stumbling out of the bar drunk the original icon will be a big dot, but if you actually interact with the event then the small dot will appear.
the only thing i can think of in that area that would be an event like that is the horse that gets loose from its owner and you chase it down and lasso it to return it.
other than that i can only think of maybe an escaped convict, but that would appear as a big dot until you actually talk to him.
