Alot of stuff is different on story mode, especially with horses. Story mode is more realistic. Your horses can die. But you can steal them and tame them and keep them and stuff like that.
Doing the mission with Uncle or Reverend Swanson will unlock Hosea's first mission - the Bear Hunt. This mission unlocks the Stable (and you can then swap horses, sell horses, buy horses, etc.). The Bear's pelt is worth decent money at the "Trapper" (not the Butcher).
Hosea's second mission is started by being in camp in the early afternoon (around 2pm) - Ms. Grimshaw will approach with news about Hosea wanting to meet you out at Emerald Ranch - this mission unlocks the Fence and also nets Arthur with some good $$

I just wanted to say I think you will like story mode. Its pretty much the same as GTA story and online mode. Story is the real thing.
