Obviously if you start from scratch everything is reset.
You'll get half as much time on your second playthrough and may not even finish it but you're right, you can get really lost in the game. You're a better player now; you'll go straight to the thing you had to find before, you won't buy stuff you don't need. You won't waste your time on unnecessary garbage. You're a gunslinger now. Be it.
This side mission is really superficial though. Nothing important that you lost by shooting him.

Yeah, but the OCD in me won't let it go! LOL I'm gonna spoil the heck out of my second play-through and read a lot of mission and side-mission walkthroughs.
Any answer as to the Rampage question? I downloaded it and got it working. I tested out some things and will probably use it for some select things like fast travel across long distances to avoid the long loading cut scenes.
