It's meant to be played after you beat the game.
For better immersion and more of an entire game over-haul, you are also meant to install/activate many other mods along side the UN2-Origins mod.

However you would need to then backup or rename the saved game files for one when switching to the other. As they will both read/write to the same folder regarding saved game progress files and the graphics settings config file.
Overall, yes it is do-able that way.
you will want to add this onto the Launch Options for RDR2 in Steam.
-skipPatcherCheck -useSteam -steamAppId=1174180 -scOfflineOnly
But then remove that line when playing RDR2 Story or Online normally.
