I am sure romance/love MAY play a role, however I'm suspicious of how big of a role that actually is as there are no indicators in the game for how it plays out. That being said, from an RP perspective, you could certainly set the flags to establish both romantic connection and how much of that there is without breaking the game or doing something which can't be undone later, like simply resetting the flags or replicating someone else's flags over existing values.
You could effectively set romance flags for your Slave Master and Gladiator in Blackthorn Arena by changing up the target IDs with the character values, although this provided no benefit as your Slave Master was never present in the arena with the Gladiator.
