Why Orpheus is a woman in this game?

Why? And does it mean, that Orpheus and her muse are lesbians in this game?

I'm pretty sure Orpheus is just a fairly effeminate man. I seem to recall Eurydice referring to her husband in the first couple of conversations I had with her.

Orpheus' speaking voice is Michael Ailshie and his singing voice is Darren Korb. Darren Korb being the guy who voices Zagreus and Skelly, and did most songs of the previous Supergiant games. So nope, just a man with a fairly effeminate appearance.

I think the way Orpheus speaks and acts fits very well with the bard-theme that he has. The Orpheus songs contain a lot of falsetto (which is an amazing technique that Darren Korb does so well) that sometimes comes across as "feminine" or at least non-gender, because falsetto-singing is usually so high-pitched that it's not quickly associated with traditional male voices. Find Top Male Falsetto's on Youtube to get a better idea of it
