So I've given Sisyphus as much nectar as I can, now I seen a lock heart symbol in the codex, but I can't give him ambrosia?

If you keep talking to him Zagreus'll eventually (or maybe he already has for you) mention the idea of lightening his sentence.

Then you have to get Hades to talk to you about it.

After that, the contractor starts getting an option to lighten Sisyphus's sentence in exchange for 10 diamonds. That is what is required to undo the lock and progress that bond further, to the point where you can give him ambrosia.

For Thanatos, it seems to be meeting him enough times during the runs (at least five?)

Meg its the same. You just gotta talk to her in the lounge enough times. Skelly you gotta get the sword's zagreus aspect to max level (takes 5 bloods total). He'll initiate a special conversation about it when you need it.
